TET Protein

TET proteins, including TET1, TET2, and TET3, are a family of dioxygenases that catalyze the demethylation of 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC), 5-formylcytosine (5fC), and 5-carboxylcytosine (5caC), facilitating DNA demethylation. These enzymes share a common core catalytic domain at their C termini. TET3 is highly expressed in oocytes and fertilized zygotes but decreases during cleavage, while TET1 and TET2 expression increases during pre-implantation development. TET1 is crucial for development and intestinal stem cell function. TET2 mutations are frequently found in hematologic malignancies, such as myelodysplastic syndrome and acute myeloid leukemia. TET3 plays a significant role in demethylation processes, including zygote formation, embryogenesis, axon regeneration, and synaptic transmission[1][2].
